「車泊」是國內目前新興的一股遊憩浪潮，本研究應用大數據文字探勘法，藉由車泊成員於臉書社群媒體中撒下的「數位麵包屑」（digital bread crumbs），以社群談論文本內容為材料，探索自2015年至2019年車泊社群整體互動情形，通過非結構化的文字，深入發掘出文字中所隱含之意義，並採取多元文字探勘技術與分析工具，適時詮釋出文本脈絡與車泊遊憩全面概況，使資料所富含的意義方得浮現。而本文使用LDA主題分析、詞語共現網絡分析、情緒分析、目的地意象分析及議題框架等方法，足夠為本研究提供不同洞見，並可歸納出一社會現象之面貌。經本研究梳理車泊社群喧嘩文本之結果發現，其社群成員主要關心的面向分別為「

遊憩體驗之分享」、「車泊地點資訊」、「車輛遊憩專門化之改裝」及「使用車泊場域之權益主張」等議題，而本研究更進一步結合社群聲量、情感分析及文字探勘技術等綜合應用，歸納出車泊成員頻繁的遊憩場域、喜好的泊點屬性與地方的關係。並以研究者自身所關心的層面，設定「環境態度與不當行爲」之議題框架，從宏觀層面進入微觀視角深入探討，發掘出車泊遊憩目前存在之問題，即是管理機關無法理解車泊行為，雖車泊社群不斷主張遊憩權益，但僅止於社群內部而以，雙方尚無法產生共識。故就車泊遊憩管理之方向，建議主管單位能夠從實務面切入，接收民間之意見回饋並制訂規範，預防遊憩衝突(recreation conflict)現象發生，綜整本

黑木耳(Auricularia auricula-judae)為可食性真菌，是東方民族常用的滋養食材，也被廣泛應用於傳統中醫藥配方。本研究主要目的在於探討黑木耳的免疫調節特性，以及分析鑑定相關的生物活性成份，以提供黑木耳醫藥保健效能之科學驗證，期促進黑木耳於保健生技產品的開發。黑木耳粉末復水後以殺菌釜高壓加熱製備為熱水萃取物(Auricularia auricula-judae hot water crude extract, AACE)，再以超過濾(ultrafiltration)技術收集不同分子量的區分物，包括：F1 (100 kDa)進行實驗。實驗內容分為二主軸：第一部分為區分物之免疫

調節效應與分子作用機制探討。第二部分為區分物之化學組成分析及活性成份鑑定。第一部分之實驗以免疫系統之先天性免疫反應第一線細胞—巨噬細胞為模式。結果顯示，黑木耳熱水萃取物具誘導巨噬細胞活化之作用，但不影響巨噬細胞對脂多醣 (lipopolysaccharide; LPS)的活化反應。AACE及其他分子量區分物單獨處理可誘導巨噬細胞活化趨向M1功能表現型，其中以F2 (10-50 kDa)的誘發功效最為顯著。各區分物之誘導強度比較結果為F2 >AACE >F3>F4>F1。活化巨噬細胞之機制主要是經由病原相關分子模式辨識受體(Pathogen-associated molecular patter

n recognition receptors; PRRs)的TLR4 (Toll-like receptor-4)細胞表面受體之訊息傳遞，誘發前發炎細胞激素釋放及M1細胞特定基因的表現。而Dectin-1受體的阻斷不會影響F2和F4對巨噬細胞的誘導活性，但會增強AACE對巨噬細胞的活化作用。以巨噬細胞及肺癌細胞共同培養為模式之實驗證實，AACE和其他區分物(F1除外)，可能藉由活化巨噬細胞而增強抗肺癌細胞增殖的功效。研究第二部分之組成份分析結果顯示，黑木耳熱水萃取物的化學成份主要為醣類(佔67.4%)，分子量主要分佈為2560 kDa、其次1134 kDa以及177 kDa。組成單醣包括：葡

萄糖酸、甘露糖、葡萄糖、木糖、鼠李糖以及N-乙醯胺基葡萄糖等主要存在於分子量高於10 kDa的區分物中。分子量小於10 kDa的區分物(F1)未檢測出甘露糖及葡萄糖酸。免疫調節活性最強的F2區分物所含葡萄糖酸、甘露糖、葡萄糖、木糖、鼠李糖和N-乙醯胺基葡萄糖之莫耳比為1.0 : 5.6 : 5.8 : 6.4 : 6.9 : 19.0。與其他區分物相較，F2含有最高莫耳比率之N-乙醯胺基葡萄糖。以核磁共振光譜分析多醣組成特性之結果顯示，F2 區分物所含多醣具有α-D-Man-(1-2)β-D-Xyl-(1-2)α-D-Rha主鏈。本研究證實Auricularia auricula-judae熱

水萃取物具有刺激免疫反應之功效，是天然免疫刺激劑 (immunostimulants) 的良好來源。最強的免疫刺組成份之分子量介於10-50 kDa，並以醣類分子為主。黑木耳多醣之免疫刺激活性可能與結構木糖、葡萄糖酸及N-乙醯胺基葡萄糖含量有關。本研究結果支持黑木耳應用作為免疫療法膳食輔佐劑的可行性。
