普通蕎麥花蜜分泌及蜜蜂授粉之研究

蕎麥 (Fagopyrum esculentum Moench) 具有糧食、保健及蜜源等功能,營養成分包含澱粉和蛋白質,富含礦物質、維生素及膳食纖維,以及芸香苷和槲皮素等機能性成分。蕎麥具自交不親和性,需仰賴授粉昆蟲授粉。本研究以蕎麥「臺中5號」為試驗材料,調查溫室裡的蕎麥開花期間每週開花數和花蜜產量變化,以及一日之中花蜜產量變化。蕎麥在種植後第50日達盛花期,最高的花蜜產量214.33 μL在種植後第43日,在盛花期前平均每朵花花蜜產量較高,通常上午花蜜產量較下午高。蜜蜂授粉試驗中對溫室中的蕎麥施以開放授粉、遮陰及完全隔離三種處理,調查蕎麥籽實的產量、一般成分及機能性成分,結果顯示不同授粉

多醣體是具有生物活性的大分子,研究已知其活性功效包含免疫調節、抗發炎及抗腫瘤等。藥用真菌在傳統醫藥的應用上已有相當悠久的歷史,其主要活性成分之一為多醣體。由於分子結構會決定多醣體的活性功效,使得多醣體的結構分析在瞭解其活性作用機制及開發具潛力之多醣體資源極為重要。然而,多醣體的結構分析面臨相當多的挑戰,包含單醣組成、序列、鍵結、首旋組態 (anomeric configuration)及分支位點之鑑定等,難以由單一方法解決所有問題。再者,傳統的分析方法為改善層析分離效率,需要對樣品進行衍生化,卻從而導致與首旋組態相關的訊息流失。本研究擬利用離子遷移質譜法(ion mobility-mass

spectrometry, IM-MS)為基礎,建立鑑定多醣體鍵結及首旋組態之方法。離子遷移質譜法在異構物(isomer)分離上具備優勢,因此本研究欲借助高解析之儀器針對醣類異構物進行分離,以取代傳統層析方法。實驗首先藉由檢視一系列三葡萄糖標準品之到達時間分布 (arrival time distribution, ATD)與斷鍵規律,建立分析方法的最佳化條件。實驗進一步的利用昆布多醣(laminarin)、糊精(dextrin)與右旋醣酐(dextran)等純化葡聚醣(glucan)進行方法驗證,結果確認此方法可成功鑑別多醣體之主鏈 結構。最後,將此方法應用於分析數種藥用真菌,包含烏靈參(X

ylaria nigripes)、舞菇(Grifola frondosa)及硫磺菇(Laetiporous sulphureus)的多醣體。結果顯示烏靈參及舞菇的水溶性非消化多醣體(water soluble non-digestible polysaccharides)由β-(1→3)-鍵結葡萄糖所聚合而成,而硫磺菇內則富含 α-(1→3)- 鍵結葡萄糖所組成之聚合物。此方法優點為樣品前處理簡單,且可快速的鑑定真菌萃取物內的水溶性非消化多醣體之結構,未來也可能應用在更多元的多醣體結構鑑定。
